To get ECM keys to autoroll you need valid EMM keys, and then yes, you need to be on the TP when new ECM keys are sent in the dvb stream.
You need to have the necessary oscam emu, configure it correctly to do autorun, without this even with valid emm keys you will not get anything.
The softcam alone may not solve the whole problem, for example an erroneous tp hash value will keep you waiting for a key.

With valid EMM's and the correct setup, and there being an ECM update broadcast in the stream, then yes new ECM's will be added to the bottom of your softcam.key file.
The file is read from the bottom up, so new keys at the bottom take priority.
The more valid EMM-keys you have in the file, the faster it is updated
